Antibodies cross-reactive with DNase1L3 and dsDNA discovered

Anti-double-stranded DNA (anti-dsDNA) antibodies, a hallmark of systemic lupus erythematosus (SLE), are a heterogeneous group of autoantibodies with differing specificities and pathogenetic effects. New research has identified a subset of autoantibodies that were previously classified as being anti-dsDNA antibodies but actually have dual reactivity to dsDNA and DNase1L3, an important enzyme in SLE pathogenesis.

“Identifying the antigenic cross-reactivity of anti-dsDNA antibodies has been of interest to investigators trying to elucidate both their potential origin and their target antigens in SLE,” says corresponding author Felipe Andrade. “This is the first study to pinpoint the source of a subset of pathogenic anti-dsDNA antibodies, which are surprisingly generated during affinity maturation of antibodies directed primarily against a self-protein antigen, namely DNase1L3.”
