Methylation inhibition and hematologic malignancies: present knowledge and future directions
Acute myelogenous leukemia and myelodysplastic syndromes are closely related disorders of the hematopoietic progenitor cells that are characterized by anemia, neutropenia and thrombocytopenia. Curative treatment is currently only available to a minority of patients and mortality is high. Exciting discoveries are allowing us to understand how the aberrant genetic and biological processes within hematopoietic progenitors give rise to these diseases. Inhibitors of the DNA methyltransferases such as azacitidine and decitabine offer great promise in the treatment of these diseases, and the authors of this supplement discuss the practical applications of these novel and exciting targeted agents.
