Peptide stapling provides control over conformation leading to improved stability and enhanced binding affinity. Non-symmetric stapling is more broadly applicable but introduces a requirement for high chemo-selectivity and site-selectivity. The past 5 years have witnessed several significant breakthroughs in addressing these challenges and are the subject of a review by Chen et al.
